Behavioral and Neurochemical Changes Induced by Oxycodone Differ Between Adolescent and Adult Mice. Yong Zhang, Roberto Picetti.
The self-administration chamber ENV-307W (21.6 cm × 17.8 cm × 12.7 cm; Med Associates, St Albans, VT) was located inside a larger box (Med Associates). The front, back, and top were constructed of 5.6 mm polycarbonate. Each chamber contained a wall with two small holes (0.9 cm diameter, 4.2 cm apart, 1.5 cm from the floor of the chamber). One hole was defined as active, the other was inactive. When the photocell in the active hole was triggered by a nose poke, the infusion pump (Med Associates) delivered an infusion of 20 μl / 3 s from a 5 ml syringe.
This study examined the effects of oral oxycodone, a prescription opioid, on several measures of impulsive behavior in healthy volunteers.
Upon arrival for each session, participants signed a compliance form stating that they had complied with the eating and drug restrictions, and provided a urine sample for pregnancy (women) and toxicology (QuikScreen, Syntron Bioresearch, Carlsbad, CA). Urines were tested for the presence of amphetamines, barbiturates, benzodiazepines, cocaine metabolites, opiates, and phencyclidine. Breath alcohol was also measured.
Of those subjects that did have evaluable data, oxycodone did not significantly alter performance on any of the impulsivity measures.

Specifically, using rats exposed to five days of relatively mild doses of oxycodone, comparable to what is prescribed to alleviate post-surgical pain in humans, researchers discovered that days after the drug exposure had ended, the rats tested in a drug-free state responded in more rigid, maladaptive ways, suggesting that impaired cognitive flexibility is an enduring consequence of oxycodone exposure.
Brief usage of the painkiller oxycodone may impair behavioral flexibility even after that use ends, suggesting impaired decision-making as an enduring consequence of exposure, according to a study conducted at the Icahn School of Medicine at Mount Sinai and published November 17 in the journal Learning and Memory.
While previous studies found that behavioral flexibility is impaired in human addicts and animals exposed to extended dosages of cocaine and amphetamine, the effects of a typical dose of oxycodone, a commonly prescribed opiate pain medication with high abuse liability, had not been systematically explored.
Oxycodone use affects an individual's physical and emotional health as well as Inappropriate, false, or hurtful words and behavior can take place and loved.
When an individual is caught up in oxycodone abuse or addiction, they tend to become consumed by their desire to use and often neglect the feelings of others, even those closest to them. While under the drug’s influence, the user may be so affected that his behavior changes drastically. He may disrespect friends, family, coworkers, or even strangers. The user may say or do things while using, that he would be ashamed to speak of sober. Inappropriate, false, or hurtful words and behavior can take place and loved ones may begin to turn away from the relationship.

So one has to take into account the dosage, the time the drug works (often the GI tract poorly absorbs drugs during headaches, and caffeine can speed things up and increase absorption efficiency), how strong an opiate receptor agonist it is, which types of opiate receptors the drug has an effect on, whether the drug is controlled release, how the individual's body metabolizes the drug, and how big a person is (the volume of blood the individual's body contains--dilution factor).
